The colon ME following chronic pressure up regulates TRPA1. DSS inflammation and chronic tension differently impacted the recruitment of LT and HT fibers and their threshold for excitation. DSS inflammation didn’t affect the composition of LT and HT fibers or the threshold of their excitation, another cause for the lack of induction of hyperalgesia by this type of inflammation. Chronic stress, however, enhanced the % of LT fibers and lowered their threshold of excitation, which may well contribute towards the boost of VMR to CRD and increase of action prospective frequency in response to CRD in LT fibers. The concurrent application of inflammation and chronic pressure showed synergistic effects: a dramatic improve of HT fibers, reduce of excitation threshold of each LT and HT fibers, and elevation of action potential frequency in response to CRD in both LT and HT fibers. The raise in the composition of HT fibers beneath concurrent inflammation and chronic stress is probably resulting from the activation of silent nociceptors (Feng and Gebhart, 2011). These synergistic effects explain the exacerbation of visceral hyperalgesia through concurrent inflammation and chronic stress and suggest similar effects in ulcerative colitis individuals (Levenstein et al., 2000a). We conclude that chronic anxiety induces visceral hypersensitivity with no inducing an inflammatory response within the colonic ME that include the nociceptors.
